New Morphinan Derivatives Exhibit Anticonvulsant Effects with Negligible Psychotomimetic Effects

Abstract

|
In order to develope novel morphinan compounds, which might possess anticonvulsant/neuroprotective properties without psychotropic effect, we synthesized eleven compounds (excluding dextromethorphan and dextrorphan) that are modified in positions 3 and 17 of the morphinan ring system. We examined protective effects of the morphinans in response to kainic acid (KA). In addition, we evaluated their locomotor behaviors in the rats. We observed that compounds 3 and 4 showed promising anticonvulsant/neuroprotective effects with behavioral safety (negligible psychotomimetic effect).
